目的 初步探讨人趋化因子样因子 1(humanchemokine likefactor 1,hCKLF 1)在系统性红斑狼疮BXSB小鼠发病中的作用。方法 利用肌肉介导的电脉冲转基因技术 ,便重组的真核表达质粒pCDI hCKLF 1在BXSB小鼠体内表达 ,并检测血清中IgM和IgG型抗DNA抗体的水平 ,尿蛋白浓度和血清尿素氮 (BUN)浓度。结果 hCKLF 1的体内表达对雌雄BXSB小鼠IgM和IgG型抗DNA抗体的产生无明显影响 ,但在短时间内显著地促进雄性BXSB小鼠尿蛋白浓度的异常升高 ,加重其肾脏炎症。结论 hCKLF 1可能是参与狼疮肾炎的介质之一 ,其效应细胞可能是单核巨噬细胞
Objective To investigate the role of human chemokine like factor 1 (hCKLF 1) in the pathogenesis of systemic lupus erythematosus BXSB mice. Methods Recombinant eukaryotic expression plasmid pCDI hCKLF 1 was expressed in BXSB mice by muscle-mediated electrical pulse gene transfection. The level of serum IgM and IgG anti-DNA antibodies, urinary protein and serum urea nitrogen BUN) concentration. Results The in vivo expression of hCKLF 1 had no significant effect on IgM and IgG anti-DNA antibody production in male and female BXSB mice, but significantly increased urinary protein concentration and increased renal inflammation in male BXSB mice in a short time. Conclusion hCKLF 1 may be involved in lupus nephritis one of the media, the effector cells may be mononuclear macrophages
