目的:探讨多巴胺D1受体激动剂SKF38393对体外培养的大鼠视皮层神经元γ氨基丁酸(GABA)-激活电流IGABA的影响。方法:选择培养11~14d的大鼠视皮层神经元,加入不同剂量的GABA,应用全细胞膜片钳技术记录IGABA并观察SKF38393对IGABA的作用。结果:视皮层神经元IGABA幅值随GABA剂量的增加而增大,且可被足量GABAA受体拮抗剂Bicuculline完全阻断。加入不同剂量SKF38393后视皮层神经元IGABA减小(P均<0.05),SKF38393剂量超过10μmol/L后产生的作用趋于稳定。联合应用10μmol/L的D1受体拮抗剂SCH23390与SKF38393后IGABA抑制率较单独应用SKF38393减小(P均<0.05)。结论:IGABA可被SKF38393抑制。多巴胺可通过D1受体作用于IGABA。
AIM: To investigate the effect of dopamine D1 receptor agonist SKF38393 on γ-aminobutyric acid (GABA) -activating current IGABA in cultured rat visual cortex. METHODS: Neurons in the visual cortex of rats were cultured for 11-14 days. Different doses of GABA were added. IGABA was recorded using whole-cell patch-clamp technique and the effect of SKF38393 on IGABA was observed. Results: The IGABA amplitude of visual cortex neurons increased with the increase of GABA dose, and could be completely blocked by sufficient GABAA receptor antagonist Bicuculline. IGABA decreased (P <0.05) in the neurons of the visual cortex after SKF38393 was added in different doses. The effect of SKF38393 at a dose exceeding 10μmol / L tended to be stable. The combined inhibition of IGABA with 10μmol / L D1 receptor antagonist SCH23390 and SKF38393 was less than that of SKF38393 alone (all P <0.05). Conclusion: IGABA can be inhibited by SKF38393. Dopamine can act on IGABA via the D1 receptor.
