目的探索铜绿假单胞菌致大鼠肺炎模型的制备方法,为后续研究奠定实验基础。方法将Sprague-Dawley(SD)大鼠48只随机分为4组:A对照组、B气管注射组、C气管插管感染组和D滴鼻感染组。适应饲喂3 d后,分别给予不同造模干预,分别于干预后5、10和15 d动态监测各亚组大鼠体重、体温、白细胞数、肺组织病理学变化等。结果各模型组大鼠行为学表现、体重、体温、白细胞、肺组织炎症病理学表现均与对照组差异有显著性;(2)各模型组均证实为铜绿假单胞菌感染,对照组则阴性。结论经滴鼻途径以铜绿假单胞菌感染大鼠,可得到合格大鼠肺炎模型;此感染途径可避免手术创口带来的炎症反应干扰,简便易行,可推广。
Objective To explore the preparation method of Pseudomonas aeruginosa induced rat pneumonia model and lay the experimental foundation for the follow-up study. Methods 48 Sprague-Dawley (SD) rats were randomly divided into 4 groups: A control group, B tracheal injection group, C tracheal intubation group and D dripping nasal infection group. After 3 days of feeding, different modeling interventions were given separately. The body weight, body temperature, white blood cell count and lung histopathological changes of rats in each subgroup were dynamically monitored at 5, 10 and 15 days after intervention respectively. Results The behavior, weight, body temperature, leukocyte and inflammatory pathological manifestations of lung tissue in each model group were significantly different from those in the control group. (2) All the model groups were confirmed as Pseudomonas aeruginosa infection while the control group negative. CONCLUSIONS: Pseudomonas aeruginosa infects rats by intranasal route to get the model of qualified rat pneumonia. This route of infection can avoid the interference of inflammatory reaction brought by surgical wounds, which is simple and easy to be promoted.
