目的建立并分析人鼻腔NK/T细胞淋巴瘤的基因表达谱。方法鼻腔NK/T细胞淋巴瘤组织和正常淋巴结组织各6例,分别进行总RNA的提取,通过逆转录过程获得标记有荧光的cDNA,分别与2张相同的包含有4096条基因的基因芯片进行杂交,根据杂交结果筛选出与鼻腔NK/T细胞淋巴瘤相关的基因。结果2张芯片中,差异表达趋势一致并且表达强度均相差1倍以上的基因共有365条(8.9%),包括癌基因和抑癌基因,细胞周期相关基因,细胞凋亡相关基因,DNA合成、修复和重组相关基因,DNA结合、转录和转录因子基因,细胞信号传递相关基因,细胞代谢相关基因以及蛋白翻译、合成相关基因等,其中37条基因差异表达4倍以上。结论鼻腔NK/T细胞淋巴瘤有多条差异表达基因,初步建立了其基因表达谱。
Objective To establish and analyze the gene expression profiles of nasal NK / T cell lymphomas. Methods Nasal NK / T cell lymphoma tissue and normal lymph node tissue were collected from 6 patients respectively. Total RNA was extracted and reversely transcribed to obtain fluorescently labeled cDNA, which were respectively compared with 2 identical gene chips containing 4096 genes Hybridization, according to the results of hybridization screening and nasal NK / T cell lymphoma-related genes. Results There were 365 genes (8.9%) in the two chips with the same trend of expression difference and more than 1 time difference in expression intensity, including oncogene and tumor suppressor gene, cell cycle related gene, apoptosis related gene, DNA Among them, 37 genes were expressed more than 4-fold in the genes of synthesis, repair and recombination, genes of DNA binding, transcription and transcription factors, genes related to cell signaling, genes related to cell metabolism, protein translation and synthesis. Conclusion There are many differentially expressed genes in nasal NK / T cell lymphomas, and their gene expression profiles are preliminarily established.
