扶正化瘀方对大鼠中毒性肾间质纤维化的作用

目的:观察扶正化瘀方对大鼠中毒性肾间质纤维化的作用。方法:Wistar大鼠40只,分为正常组、模型组、扶正化瘀方药物组。采用二甲基亚硝胺(dimethylnitlrosamine,DMN)腹腔注射4周和含汞等重金属的饲料喂饲造模8周,制备大鼠肝纤维化-肾间质纤维化模型。药物组以扶正化瘀方自第5周起灌胃,1次/d,共4周。肾组织HE染色及六胺银(periodicacid-silvermetheramine,PASM)染色,观察肾组织炎症与胶原纤维沉积变化;试剂盒方法检测血清尿素氮与肌酐含量,观察肾功能变化;盐酸水解法测定肾组织羟脯氨酸(hydroxyproline,Hyp)含量,观察胶原生成变化。结果:模型大鼠肾小管上皮细胞浊肿、坏死与脱落,炎性细胞浸润;肾间质增宽,大量胶原沉积;肾小管基底膜增厚,出现皱褶;血清尿素氮与肌酐含量上升,肾组织Hyp含量显著增加。扶正化瘀方药物组明显减轻模型大鼠肾小管炎症坏死与肾间质胶原沉积,减轻肾组织Hyp含量,降低异常增高的血清尿素氮与肌酐水平。结论:DMN腹腔注射与汞等重金属饲料口服可造成大鼠肾间质纤维化,扶正化瘀方可明显改善大鼠肾功能,有抗肾间质纤维化的作用。 Objective: To observe the effect of Fuzheng Huayu Recipe on toxic renal interstitial fibrosis in rats. Methods: Forty Wistar rats were divided into normal group, model group and Fuzheng Huayu Recipe group. Animal model of hepatic fibrosis-renal interstitial fibrosis was prepared by intraperitoneal injection of dimethylnitramine (DMN) for 4 weeks and feed containing mercury and other heavy metals for 8 weeks. The drug group took Fuqi Huatan Fangfang from the 5th week, once a day for 4 weeks. Renal tissue HE staining and periodic acid-silvermetheramine (PASM) staining were used to observe the changes of renal inflammation and collagen fiber deposition. Serum urea nitrogen and creatinine levels were measured by kit method, and renal function was observed. Hydrochloric acid was used to determine renal tissue hydroxylation. Hydroxyproline (Hyp) content was observed for changes in collagen production. RESULTS: Tubular epithelial cells in the model rats had turbid swelling, necrosis and shedding, inflammatory cell infiltration, broadening of the renal interstitial substance, and deposition of a large amount of collagen; thickening of the basement membrane of the renal tubules and appearance of folds; increased levels of serum urea nitrogen and creatinine. The renal tissue Hyp content increased significantly. Fuzheng Huayu Recipe group significantly reduced tubular necrosis and necrosis and renal interstitial collagen deposition, reduced renal Hyp content, and decreased abnormally elevated serum urea nitrogen and creatinine levels. Conclusion: DMN intraperitoneal injection and mercury and other heavy metals feed can cause kidney interstitial fibrosis in rats. Fuzheng Huayu Decoction can obviously improve the renal function of rats, and it has anti-renal interstitial fibrosis.
