目的探讨XLOC_004122在结核患者支气管肺泡灌洗液的表达以及功能,评价长链非编码RNA(XLOC_004122)作为诊断MTB感染新标志物的潜力。方法收集解放军第309医院2014年10月-2015年3月结核科48例患者支气管肺泡灌洗液标本,结合患者的临床症状、体征、实验室检查、影像学检查等将其分为结核病组和非结核对照组,应用实时荧光定量PCR检测XLOC_004122的差异表达;通过基因GO功能注释、富集分析和KEGG信号转导通路富集分析,阐明XLOC_004122靶基因参与调控的细胞功能与信号通路。结果与非结核对照组相比,结核病组XLOC_004122的表达量显著下调(t=-2.487,P<0.01);GO分析结果显示XLOC_004122靶基因功能主要富集于离子通道的活性、通道复合物的合成、跨膜转运活性等过程;KEGG信号转导通路主要富集于Ca2+、toll样受体等信号通路,通过XLOC_004122共表达的Ca2+通道信号通路可以看出其在NCX、OPCR、CaV1、PTK、MLCK和CAMK中均有差异表达。结论 XLOC_004122在结核感染中呈相对低表达,且可能与结核病的发生有关,可尝试将其作为诊断结核感染的新指标。
Objective To investigate the expression and function of XLOC_004122 in bronchoalveolar lavage fluid of patients with tuberculosis and to evaluate the potential of long chain noncoding RNA (XLOC_004122) as a new marker for the diagnosis of MTB infection. Methods The samples of bronchoalveolar lavage fluid from 48 patients with tuberculosis in the 309th Hospital of PLA from October 2014 to March 2015 were collected and divided into tuberculosis group and tuberculosis group according to clinical symptoms, signs, laboratory tests and imaging examination. Non-tuberculosis control group, real-time fluorescent quantitative PCR was used to detect the differential expression of XLOC_004122. The function and signal pathway of XLOC_004122 target gene were clarified through gene GO annotation, enrichment analysis and enrichment analysis of KEGG signal transduction pathway. Results The expression of XLOC_004122 in tuberculosis patients was significantly down-regulated (t = -2.487, P <0.01) compared with non-tuberculosis controls. The results of GO analysis showed that the function of XLOC_004122 target genes was mainly enriched in the activity of ion channels and the synthesis of channel complexes , Transmembrane transport activity and other processes; KEGG signal transduction pathway is mainly enriched in Ca2 +, toll-like receptors and other signaling pathways, through XLOC_004122 co-expression of Ca2 + channel signaling pathway can be seen in NCX, OPCR, CaV1, PTK, MLCK And CAMK are differentially expressed. Conclusion XLOC_004122 is relatively low expression in tuberculosis infection and may be related to the occurrence of tuberculosis. It may be used as a new indicator to diagnose tuberculosis infection.
