目的:测定秃叶黄皮树和黄皮树中黄柏碱、木兰花碱、巴马汀、小檗碱、黄柏酮的含量,并比较不同品种黄柏药材有效成分含量的差异。方法:采用HPLC,Xtimate C18色谱柱(4.6 mm×250 mm,5μm),流动相乙腈(A)-0.1%磷酸水溶液(B),梯度洗脱,检测波长220 nm,流速1.0 mL·min-1,柱温30℃。结果:5种成分在上述条件下分离良好。黄柏碱、木兰花碱、巴马汀、小檗碱、黄柏酮分别在0.001 7~0.07,0.002~0.08,0.000 5~0.02,0.01~0.4,0.000 5~0.02 g·L-1呈良好线性关系,r分别为0.999 9,0.999 9,0.999 9,0.999 9,0.999 5,平均回收率均>97%,(RSD<3%,n=9)。结论:该方法简便、准确、重复性好,可作为秃叶黄皮树与黄皮树质量控制的方法。秃叶黄皮树中黄柏碱、木兰花碱、巴马汀、小檗碱、黄柏酮含量与黄皮树无明显差异;且已成为黄柏药材的主流商品,建议将秃叶黄皮树与黄皮树一同作为黄柏收入药典。
OBJECTIVE: To determine the content of cork base, magnolol, palmatine, berberine, and berberine in the leaves of Huangcao and Huangpi, and to compare the differences of the active ingredients in the different varieties of Huangpu. Methods: The mobile phase consisted of acetonitrile (A) -0.1% phosphoric acid (B) and gradient elution on a Xtimate C18 column (4.6 mm × 250 mm, 5 μm) with a detection wavelength of 220 nm and a flow rate of 1.0 mL · min -1 The column temperature was 30 ℃. Results: The five components were well separated under the above conditions. Phellodendron, magnolol, palmatine, berberine and phellodendron showed a good linear relationship between 0.001 7 ~ 0.07,0.002 ~ 0.08,0.000 5 ~ 0.02,0.01 ~ 0.4,0.000 5 ~ 0.02 g · L-1 , r were 0.999 9,0.999 9,0.999 9,0.999 9,0.999 5, the average recovery was> 97% (RSD <3%, n = 9). Conclusion: The method is simple, accurate and reproducible. It can be used as a quality control method for the leafless leaves and the yellow-bark tree.
