目的观察胆宁片对非酒精性脂肪肝(NAFLD)大鼠琥珀酸脱氢酶(SDH)表达的影响。方法采用高脂饲料喂养的方式复制非酒精性脂肪肝大鼠模型,同时给予胆宁片、多烯磷脂酰胆碱、绞股蓝总甙片进行治疗,观察大鼠肝脏病理形体学的变化,血清甘油三酯(TG),总胆固醇(TC),高、低密度脂蛋白(HDL-C、LDL-C)的动态变化,应用分光光度计法观察大鼠肝脏SDH的表达情况。结果与正常组比较,模型组大鼠肝脏体积及重量均增大,病理切片见细胞质内存在大小不一脂肪空泡,细胞核受到不同程度挤压,部分细胞出现坏死,TG、TC均明显升高,HDL-C下降,SDH含量显著下降(P<0.01)。与模型组比较,胆宁片能显著减轻肝脏脂肪变性(P<0.05),SHD含量显著升高(P<0.01)。结论胆宁片可以提高NAFLD大鼠肝脏SDH含量,从而达到治疗NAFLD的作用。
Objective To observe the effect of Danning Tablet on the expression of succinate dehydrogenase (SDH) in nonalcoholic fatty liver disease (NAFLD) rats. Methods The model of nonalcoholic fatty liver was reproduced by feeding with high fat diet. At the same time, Danning tablets, polyene phosphatidylcholine and Gynostemma pentaphylla tablets were treated. The changes of hepatic pathomorphology, serum glycerol Triglyceride (TG), total cholesterol (TC), high and low density lipoprotein (HDL-C, LDL-C) were measured. The expression of SDH in rat liver was observed by spectrophotometer. Results Compared with the normal group, the liver volume and weight of rats in the model group were all increased. Pathological sections showed that there were different sizes of vacuolar vacuoles in the cytoplasm. The nuclei were squeezed to some extent and some cells were necrotic. TG and TC were significantly increased , HDL-C decreased, SDH content decreased significantly (P <0.01). Compared with the model group, Danning Tablet can significantly reduce hepatic steatosis (P <0.05), SHD content was significantly increased (P <0.01). Conclusion Danning Tablet can improve the NAFLD rat liver SDH content, so as to achieve the effect of NAFLD.
