目的 :探讨中药复方益气解毒颗粒对人胚鼻咽上皮细胞转化的阻抑作用。方法 :二亚硝基哌 (DNP)诱导人胚鼻咽上皮细胞转化 ,同时用益气解毒颗粒作用于DNP诱导的鼻咽上皮细胞 ,共同培养 ,然后收集不同代数细胞 ,酶连免疫聚合酶链反应 (PCR -ELISA)和巢式聚合酶链反应 (Nested -RT -PCR)法检测实验细胞端粒酶活性和端粒酶RNA的表达。结果 :加益气解毒颗粒药液组鼻咽细胞形态明显好于盐水对照组 ,其端粒酶活性 (0 34± 0 19)也明显低于对照组 (0 73± 0 2 7) (P <0 0 5 )。结论 :益气解毒颗粒能够抑制鼻咽上皮细胞端粒酶的激活 ,进而阻断DNP诱导的细胞转化。
Objective: To investigate the inhibitory effect of Yiqi Jiedu Granules on the transformation of human embryonic nasopharyngeal epithelial cells. METHODS: Dinitroso piperazine (DNP) was used to induce transformation of human embryonic nasopharyngeal epithelial cells. At the same time, DNP-induced nasopharyngeal epithelial cells were treated with Yiqi Jiedu Granules. The cells were co-cultured and then collected for different generations of cells. The enzyme-linked immuno-polymerase chain reaction was performed. The telomerase activity and telomerase RNA expression were detected by PCR-ELISA and nested-RT-PCR. Results: The morphology of nasopharyngeal cells in Jiayiqijiedu Granule group was significantly better than that in the saline control group, and the telomerase activity (0 34 ± 0 19) was also significantly lower than that of the control group (0 73 ± 0 2 7) (P < 0 0 5 ). Conclusion : Yiqi Jiedu Granules can inhibit the activation of telomerase in nasopharyngeal epithelial cells, which in turn blocks DNP-induced cell transformation.
