目的:观察益肾软坚之二甲丸对大鼠增生型子宫内膜中雌激素受体(ER)、孕激素受体(PR)以及碱性成纤维细胞生长因子(BFGF)、血管内皮生长因子(VEGF)表达的影响,探讨其治疗功能性子宫出血之子宫内膜增生的作用机制。方法:将健康SD大鼠80只随机分为空白组、模型组和二甲丸高、中、低剂量组及西药组,其中模型组和西药组各14只,其余每组各13只。空白组常规饲养,其余组均行去势术,术后除空白组外均给予补佳乐灌胃建立大鼠子宫内膜增生模型。空白组与模型组仅予等量蒸馏水灌胃,二甲丸高、中、低剂量组分别给予相应剂量二甲丸汤剂灌胃,西药组给予地屈孕酮研磨灌胃,均连续灌胃14d。采用免疫组化SP法检测各组大鼠子宫内膜中ER、PR、BFGF、VEGF的表达水平。结果:通过干预后二甲丸各组BFGF、VEGF均明显下降;相关分析发现,治疗前后比较BFGF、VEGF与ER呈显著正相关,而与PR则呈显著负相关,这种变化与正常大鼠子宫内膜相符合。结论:益肾软坚法可能通过ER、PR的介导作用,降低BFGF、VEGF的表达,进而抑制血管过度增殖,从而达到阻断子宫内膜异常增生的目的。
Objective: To observe the effects of Yishen Ruanjian Erjia Pills on the expression of estrogen receptor (ER), progesterone receptor (PR), basic fibroblast growth factor (BFGF), vascular endothelial growth factor (VEGF) expression in the treatment of functional uterine bleeding endometrial hyperplasia mechanism. Methods: Eighty healthy SD rats were randomly divided into two groups at random: model group, high, medium and low dose dimethylphenidate group and western medicine group. There were 14 rats in model group and western medicine group, and 13 rats in each group. The rats in the blank group were routinely housed, and the other groups underwent castration. After the operation, all rats except the blank group were given gavage to establish a model of endometrial hyperplasia in rats. The blank group and the model group were given only the same amount of distilled water gavage, high dose group, middle and low dose group were given the corresponding dose of Xiaoyao decoction gavage, 14d. Immunohistochemical SP method was used to detect the expression of ER, PR, BFGF and VEGF in endometrium of rats in each group. Results: BFGF and VEGF in each group decreased significantly after intervention. Correlation analysis showed that there was a significant positive correlation between BFGF, VEGF and ER before and after treatment, but negatively correlated with PR, which was significantly different from that in normal rats Endometrium consistent. Conclusion: Yishenjianjian may reduce the expression of BFGF and VEGF through the mediation of ER and PR, and then inhibit the hyperproliferation of blood vessels, so as to achieve the goal of blocking the abnormal proliferation of endometrium.
