目的探讨糖皮质激素对哮喘小鼠肺组织水通道蛋白5(AQP5)表达的影响。方法45只清洁级BALB/c小鼠随机分为对照组、哮喘组和地塞米松干预组,每组15只。采用免疫组化法检测肺组织中AQP5蛋白的表达。结果哮喘组肺组织AQP5蛋白表达较对照组明显减低[(0.1186±0.0007)vs.(0.2060±0.0005)](P<0.01),地塞米松干预后AQP5蛋白表达较哮喘组增加[(0.1502±0.0008)vs.(0.1186±0.0007)](P<0.01)。结论哮喘小鼠肺组织AQP5蛋白表达下调,AQP5可能参与了哮喘发病时气道黏液分泌的调节,糖皮质激素可上调哮喘小鼠肺组织AQP5蛋白的表达。
Objective To investigate the effect of glucocorticoids on the expression of aquaporin 5 (AQP5) in the lungs of asthmatic mice. Methods Forty-five clean BALB / c mice were randomly divided into control group, asthma group and dexamethasone intervention group, 15 rats in each group. The expression of AQP5 protein in lung tissue was detected by immunohistochemistry. Results Compared with control group, the expression of AQP5 in asthmatic group was significantly lower than that in control group [(0.1186 ± 0.0007) vs. (0.2060 ± 0.0005)], P <0.01. The expression of AQP5 in asthmatic group was significantly increased than that in asthma group [(0.1502 ± 0.0008) ) vs. (0.1186 ± 0.0007)] (P <0.01). Conclusions The expression of AQP5 protein in the lung tissue of asthmatic mice is down-regulated. AQP5 may be involved in the regulation of airway mucus secretion in asthmatic mice. Glucocorticosteroid may up-regulate the expression of AQP5 in lung tissue of asthmatic mice.
