目的:建立盾叶薯蓣中伪原薯蓣皂苷的含量测定方法。方法:采用高效液相色谱-蒸发光散射检测法(HPLC-ELSD),Dikma C18色谱柱(4.6 mm×250 mm,5μm),流动相乙腈-水(30∶70),流速1.0 mL.min-1;蒸发光散射检测器参数:漂移管温度60℃,气压4.5 kPa。结果:伪原薯蓣皂苷在10~40μg(r=0.999 8)线性关系良好,平均回收率98.7%(RSD 1.6%,n=6)。结论:该方法简便、快速、灵敏、准确,可作为盾叶薯蓣的质量控制方法。
Objective: To establish a method for the determination of pseudo-yam saponin in Dioscorea zingiberensis. Methods: HPLC-ELSD was used on a Dikma C18 column (4.6 mm × 250 mm, 5 μm). The mobile phase consisted of acetonitrile-water (30:70), flow rate 1.0 mL · min- 1; evaporative light scattering detector parameters: drift tube temperature 60 ℃, pressure 4.5 kPa. Results: The pseudo - original diosgenin showed good linearity at 10 ~ 40μg (r = 0.999 8) with an average recovery of 98.7% (RSD 1.6%, n = 6). Conclusion: The method is simple, rapid, sensitive and accurate and can be used as a quality control method for Dioscorea zingiberensis.
