目的:对冬虫夏草虫体和子座部分的甾醇和核苷类化合物进行多组分定性定量分析,并比较核苷类化合物的提取方法。方法:用HPLCDAD联用色谱检测,借助HELP法解析,基于组分的色谱保留时间和光谱曲线进行定性,然后用总体积积分法定量。结果:HPLCDAD联用色谱检测和HELP法解析相结合,可验证色谱峰的纯度,分辨重叠峰,得到各组分的纯色谱和光谱曲线。该法的样品回收率为9997%~10081%,虫体和子座部分的化学成分存在差异,用水提取核苷类化合物的效果较好。结论:综合利用色谱和光谱两方面信息,可提高组分定性定量结果的准确度。
OBJECTIVE: To qualitatively and quantitatively analyze the components of sterols and nucleosides of Cordyceps militaris and sub-locus, and to compare the methods of extracting nucleosides. Methods: HPLC-DAD coupled with chromatography, with HELP analysis, based on the chromatographic retention time of the components and spectral curves were qualitative, and then quantified using the total volume integral method. Results: The HPLC-DAD combined with HELP method can verify the purity of chromatographic peaks and distinguish the overlapping peaks. The pure chromatograms and spectral curves of each component were obtained. The recovery rate of the method was 9997% -10081%. There were differences in the chemical composition between the parasites and the sub-locus. The effect of extracting the nucleosides with water was better. Conclusion: Comprehensive utilization of both chromatographic and spectral information can improve the accuracy of quantitative qualitative and quantitative results of components.
