目的:通过观察活血消瘿方对结节性甲状腺肿模型大鼠增殖细胞核抗原(PCNA)表达以及甲状腺肿细胞凋亡水平的影响,探讨活血消瘿方治疗结节性甲状腺肿可能的作用机制。方法:分别采用Western Blot法检测PCNA在Wistar大鼠甲状腺组织中的表达及采用流式细胞仪Annexin V/PI双染色法检测Wistar大鼠甲状腺肿组织中的细胞凋亡水平。结果:LT4和大、中、小剂量活血消瘿方均可降低模型大鼠甲状腺细胞PCNA的表达及促进大鼠甲状腺细胞的凋亡,并且小剂量活血消瘿方作用最为显著。结论:活血消瘿方治疗结节性甲状腺肿可能是通过抑制甲状腺肿细胞的增殖及促进其凋亡。
OBJECTIVE: To observe the effect of Huoxue Xiaopan Recipe on the expression of proliferating cell nuclear antigen (PCNA) and the apoptosis of goiter in nodular goiter model rats, and to explore the possible mechanism of Huoxue Xiaopan Recipe in treating nodular goiter. Methods: The expression of PCNA in Wistar rat thyroid tissue was detected by Western Blot and the apoptosis in goiter of Wistar rats was detected by flow cytometry Annexin V / PI double staining. Results: LT4 and large, medium and small doses of Huoxue Xiaopan can reduce the expression of PCNA in rat thyroid cells and promote the apoptosis of rat thyrocytes, and the effect of Xiaoer Huoxue Xiaopan is the most obvious. Conclusion: The treatment of nodular goiter by Huoxue Xiaopan decoction may inhibit the proliferation and promote the apoptosis of goiter cells.
