目的 建立测定鞣苦胶囊中苦参总生物碱含量的方法。方法 采用溶剂萃取酸性染料比色法测定苦参总生物碱含量 ,以氧化苦参碱为对照品 ,溴麝香草酚蓝为酸性染料 ,在pH 7 6缓冲溶液条件下 ,用氯仿萃取 ,检测波长 4 17nm。结果 氧化苦参碱在 0 6 98~ 13 96mg L范围内与吸光度呈良好线性关系 ,回归方程为Y =2 0 2 4 3X - 1 5 136 ,r =0 9993(n =5 ) ,氧化苦参碱的平均回收率为 10 2 6 7% ,RSD =0 74 % (n =5 )。结论 所建立方法简便、准确、专属性强。可用于鞣苦胶囊中苦参总生物碱含量测定。
Objective To establish a method for the determination of total alkaloids in Sophora flavescens Ait. Methods The content of total alkaloids in Sophora flavescens was determined by solvent extraction acid dye colorimetric method. Oxymatrine was used as the reference substance and bromothymol blue was the acid dye. The extraction was performed with chloroform at pH 7.6 buffer solution. 4 17nm. Results Oxymatrine showed a good linear relationship with the absorbance in the range of 0 6 98 ~ 13 96 mg L. The regression equation was Y = 2 0 2 4 3X - 1 5 136, r = 9993 (n = 5). The average recovery of the alkali was 10 2 6 7%, and the RSD was 0.74% (n = 5). Conclusion The established method is simple, accurate and specific. It can be used to determine the content of total alkaloids in Sophora flavescens Ait.
