目的探讨血府逐瘀汤对人脐静脉内皮细胞(HUVEC)分泌血管内皮生长因子(VEGF)、血小板源性生长因子(PDGF)的影响及其与促进HUVEC增殖的关系。方法采用血府逐瘀汤含药血清体外干预HUVEC培养,通过MTT法和酶联免疫吸附分析(ELISA)法观察血府逐瘀汤对HUVEC的体外增殖和分泌VEGF、PDGF的影响作用。结果MTT检测结果显示血府逐瘀汤含药血清浓度为10%时对HUVEC具有显著增殖作用;ELISA检测结果显示血府逐瘀汤具有促进HUVEC分泌VEGF的作用,而对PDGF的分泌无明显的作用。结论血府逐瘀汤在一定剂量范围内能促进HUVEC增殖和促进HUVEC分泌VEGF,而对PDGF的分泌无明显促进作用;提示血府逐瘀汤可能通过促进HUVEC分泌VEGF而促进细胞增殖,从而起到祛瘀生新的作用。
Objective To investigate the effect of Xuefu Zhuyu Decoction on the secretion of vascular endothelial growth factor (VEGF) and platelet-derived growth factor (PDGF) in human umbilical vein endothelial cells (HUVEC) and its relationship with the proliferation of HUVEC. Methods The serum containing Xuefu Zhuyu decoction was used to intervene HUVEC culture in vitro. The effects of Xuefu Zhuyu decoction on the proliferation of HUVEC and the secretion of VEGF and PDGF were observed by MTT assay and enzyme-linked immunosorbent assay (ELISA). Results The results of MTT assay showed that the serum concentration of Xuefu Zhuyu Decoction had a significant proliferative effect on HUVEC. The results of ELISA showed that Xuefu Zhuyu Decoction had the effect of promoting the secretion of VEGF by HUVEC, but had no obvious effect on the secretion of PDGF. effect. Conclusion Xuefu Zhuyu Decoction can promote the proliferation of HUVEC and promote the secretion of VEGF in HUVEC within a certain dose range, but has no significant effect on the secretion of PDGF.
