虽已有研究证明7,8-二羟基黄酮[7,8-dihydroxyflavone,(7,8-DHF)]具有有效的神经保护作用,但其对血管功能的作用还不是很明确。方法:通过从正常大鼠分离的主动脉环,检测7,8-DHF对由肾上腺素诱导的动脉收缩的影响。应用阻断剂、Western blot和原代培养血管平滑肌细胞研究其影响机制。用尾套法测量大鼠的血压。结果:7,8-DHF剂量依赖性扩张肾上腺素预收缩的内皮细胞完整的主动脉环,50%最大效应(EC50)浓度约24μmol/L。左旋硝基精氨酸甲酯盐酸盐(一氧化氮合酶抑制剂)和ODQ(1H-[1,2,4]oxadiazolo[4,3-a]quinoxalin-1-one,可溶性鸟苷酸环化酶受体阻滞剂)均显著降低7,8-DHF的
Although studies have shown that 7,8-dihydroxyflavone (7,8-DHF) has an effective neuroprotective effect, its effect on vascular function has not been clarified yet. Methods: The effect of 7,8-DHF on epinephrine induced arterial contractions was examined by aortic rings isolated from normal rats. Blocking agents, Western blot and primary cultured vascular smooth muscle cells were used to study the mechanism. Rat tail blood pressure was measured by tail cuff method. RESULTS: 7,8-DHF dose-dependently dilated the intact aortic rings of epinephrine precontracted endothelial cells with a 50% maximal concentration (EC50) of about 24 μmol / L.
