目的:观察黄芩茎叶总黄酮(Scutellaria baiculensis stem-leaf total flavonoid,SSTF)抑制大鼠心肌缺血再灌注时细胞凋亡及可能机制。方法:40只SD大鼠随机分为假手术组(sham operation group,SHAM组)、缺血再灌注组(Ischemia reperfusion group,IR组)、SSTF 3个剂量组。SSTF组于术前1周ig不同剂量的SSTF(17.5,35,70 mg.kg-1.d-1),另2组ig给等量生理盐水。采用结扎大鼠左冠状动脉前降支方法制备心肌缺血再灌注模型。缺血30 min,再灌注2 h后,采用Annexinv/PI双染,流式细胞仪检测心肌细胞的凋亡情况,RT-PCR检测caspase-3基因mRNA的表达,采用蛋白印记法定量检测心肌细胞caspase-3基因的表达情况。结果:与SHAM组相比,IR组的心肌细胞的凋亡率明显增加(P<0.01),并且caspase-3基因的mRNA和蛋白表达均增加(P<0.01);与IR组相比,SSTF可明显降低心肌细胞的凋亡率(P<0.05,P<0.01);降低caspase-3基因的mRNA和蛋白表达(P<0.05,P<0.01)。结论:SSTF抑制心肌细胞凋亡可能与降低caspase-3基因的mRNA和蛋白表达有关。
OBJECTIVE: To observe the effect of Scutellaria baiculensis stem-leaf total flavonoid (SSTF) on apoptosis and its possible mechanism during myocardial ischemia-reperfusion in rats. Methods: Forty SD rats were randomly divided into sham operation group (SHAM group), Ischemia reperfusion group (IR group) and SSTF three dose groups. The SSTF group had different doses of SSTF (17.5, 35, 70 mg.kg-1.d-1) one week before surgery, and the other two groups had the same amount of saline. Myocardial ischemia-reperfusion model was established by ligating left anterior descending coronary artery in rats. The apoptosis of cardiomyocytes was detected by flow cytometry, the expression of caspase-3 mRNA was detected by RT-PCR and the expression of caspase-3 mRNA was detected by Western blot.2. caspase-3 gene expression. Results Compared with SHAM group, the apoptosis rate of cardiomyocytes in IR group increased significantly (P <0.01), and the mRNA and protein expression of caspase-3 gene increased (P <0.01). Compared with IR group, SSTF (P <0.05, P <0.01), and decreased the mRNA and protein expression of caspase-3 (P <0.05, P <0.01). Conclusion: Inhibition of SSTF on cardiomyocyte apoptosis may be related to the decrease of caspase-3 mRNA and protein expression.
