多种瘤抑制基因(MTS1)表达产物P16蛋白作为细胞周期依赖性蛋白激酶(CDK)的抑制物,使Rb蛋白磷酸化受阻,细胞不能由G_1期进入S期,进而抑制细胞分裂增殖,为进一步开展消化道肿瘤防治研究,我们构建了MTS1真核表达载体,现报告如下。 1 材料和方法 1.1 材料 pUC19-P16质粒和pcDNA3真核表达载体由第四军医大学生化教研室提供,Plasmid purification kit购自上海华顺公司,PCR purification kit购自美国Clontech公司。BamHI,EcoRI,T_4 DNA ligase,Xba Ⅰ,Hind Ⅲ等限制酶,DNA Marker及其他常用试剂购自美国Promega公司和中国华美公
P16 protein, a tumor suppressor gene (MTS1) expression product, acts as an inhibitor of cell cycle-dependent protein kinase (CDK), blocking the phosphorylation of Rb protein, and preventing cells from entering the S phase from G1 phase, thereby inhibiting cell division and proliferation. To carry out research on the prevention and control of digestive tract tumors, we have constructed an MTS1 eukaryotic expression vector, which is reported below. 1 Materials and methods 1.1 Materials pUC19-P16 plasmid and pcDNA3 eukaryotic expression vector were provided by the Fourth Military Medical University, Plasma purification kit was purchased from Shanghai Huashun Company, and PCR purification kit was purchased from Clontech, USA. Restriction enzymes such as BamHI, EcoRI, T_4 DNA ligase, Xba I, Hind III, DNA Marker, and other commonly used reagents were purchased from Promega Corporation of the United States and China
