本文报道了杂交水稻12个不同组合幼穗的体细胞胚诱导与同步化的研究结果。结果表明:材料的基因型对其体细胞胚发生有明显的影响,植物激素2,4—D在杂交水稻体细胞胚胎发生与发育中起重要作用,BA则起一定的协调促进作用。通过研究建立了杂交水稻悬浮细胞系与产生大量体细胞胚的培养程序。利用生长素2,4—D结合机械筛选的方法,实现体细胞胚同步率达85%以上。通过组织学观察发现体细胞胚的发生基本来源于胚性愈伤组织表面的细胞,其发育似合子胚,但出现了双子叶形胚和少数畸形胚。
In this paper, we report the results of the somatic embryo induction and synchronization in 12 young ears of hybrid rice. The results showed that the genotype of the plant had a significant effect on the somatic embryogenesis. The plant hormone 2,4-D played an important role in the somatic embryogenesis and development of hybrid rice and BA played a coordinating role. The establishment of a hybrid rice suspension cell line and produce a large number of somatic embryos culture program. Using auxin 2,4-D combined with mechanical screening method, somatic embryos synchronization rate of 85% or more. Histological observation revealed that the occurrence of somatic embryos basically originated from cells on the surface of embryogenic callus and developed zygotic embryos. However, there appeared dicotyledonous embryos and a few abnormal embryos.
