目的 观察波形纤维蛋白在老年性白内障晶状体上皮细胞的变化。方法 用卵白素 生物素过氧化物酶法对 2 2例老年性白内障患者晶状体前囊膜上皮细胞进行波形纤维蛋白染色 ,采用包埋前免疫酶电镜技术处理 6个晶状体前囊膜标本 ,并观察其超微结构 ;利用十二烷基硫酸钠聚丙烯酰胺凝胶电泳及Westernblot法分析 4个晶状体表层组织 (前囊膜、上皮细胞和表层皮质 )中的波形纤维蛋白。结果 老年性白内障晶状体上皮细胞的波形纤维蛋白表达减弱 ,与对照组比较差异有显著性 (t=2 0 948,P <0 0 5 ) ;其纤维分布稀疏 ,聚集成束 ,细胞器变性 ;未成熟期白内障晶状体可检测到波形纤维蛋白 ,成熟期白内障晶状体的波形纤维蛋白含量极少。结论 老年性白内障晶状体上皮细胞及表层皮质中波形纤维蛋白的减少 ,与各种因素引起的波形纤维蛋白降解有关 ,可能是晶状体中细胞结构和功能破坏的表现 ,是白内障形成的重要机制。
Objective To observe the changes of corrugated fibrin in lens epithelial cells of senile cataract. Methods The anterior lens epithelial cells of 22 patients with age-related cataract were stained with the fibrinogen by avidin biotin peroxidase method, and 6 anterior lens capsule samples were treated by immunoenzymatic electron microscopy Their ultrastructures were observed. Waveform fibrils in four lens epithelial tissues (anterior capsule, epithelial cells and superficial cortex) were analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis and Western blotting. Results Compared with the control group, the expression of vimentin in the lens epithelial cells in aged patients with cataract decreased significantly (t = 2 0 948, P <0 05). The fiber distribution was sparse, clustered into bundles and organelles were degenerated. Phased cataract lens can detect the waveform of fibrin, mature cataract lens waveform fibrin content is minimal. Conclusions The decrease of waveform fibrin in lens epithelial cells and superficial cortex of senile cataract may be related to the degradation of waveform fibrin induced by various factors, which may be the manifestation of cell structure and function destruction in lens. It is an important mechanism of cataract formation.
