以发酵产鼠李糖脂的铜绿假单胞菌Z-2为发酵菌株,对造纸综合废水作为发酵用水发酵产生物表面活性剂进行了研究。结果表明:综合废水在添加炸货油和酵母提取物后可以发酵产生鼠李糖脂,表面张力降低50%以上。单因素摇瓶实验初步优化造纸综合废水培养基碳氮源表明:炸货油发酵效果好于甘油,粗甘油不能被利用,较适合添加的氮源为尿素,但随着用量增大,发酵液表面活性反而下降。利用Design Expert 8.0.6软件对废水发酵培养基通过Box-Behnken设计(BBD)进行响应面优化,得到最佳培养基的各成分最优水平为:炸货油2.64%、尿素0.52g·L~(-1)、氯化钾0.08 g·L~(-1)。在5 L发酵罐中一次性加入碳源的情况下,鼠李糖脂产量达5.56 g·L~(-1)。
Fermentation of rhamnolipid-producing Pseudomonas aeruginosa Z-2 was used as the fermentation strain to study the comprehensive utilization of papermaking wastewater as fermentative surfactant for fermentation. The results showed that the synthetic waste water could produce rhamnolipid by fermentation after adding oil and yeast extract, and the surface tension decreased by more than 50%. Single-factor shake flask experiment preliminary optimization of carbon and nitrogen sources in comprehensive wastewater treatment of papermaking wastewater showed that the fermentation effect of the crude oil was better than that of glycerol, the crude glycerol could not be utilized, and the more suitable nitrogen source was urea. However, with the increase of the amount, The surface activity decreased. Optimization of the response surface of the wastewater fermentation medium through the Box-Behnken design (BBD) using Design Expert 8.0.6 software gave the optimum levels of optimal components of the fermentation medium: 2.64% of the crude oil, 0.52g · L ~ (-1) and potassium chloride 0.08 g · L -1. The rhamnolipid yield reached 5.56 g · L -1 with one-time carbon source addition in a 5 L fermentor.
