蚕丝由于其出色的材料性能,引起人们对蚕丝基础及应用研究的广泛关注,其中丝蛋白与金属离子的相互作用一直是人们研究的热点.利用远紫外圆二色谱(circulardichroism,CD)方法研究了水溶液中金属离子Cu(Ⅱ)对蚕丝素蛋白构象的影响.结果表明,一定量Cu(Ⅱ)离子的引入有利于丝素蛋白分子链从无规卷曲向β-折叠构象的转变,然而Cu(Ⅱ)离子的进一步增加反而会抑制β-折叠构象的形成.同时研究结果还表明,Cu(Ⅱ)离子对丝素蛋白构象的影响遵循“成核依赖性”机理.这一机理与存在于中枢神经系统中的朊蛋白在Cu(Ⅱ)离子诱导下的病变过程非常类似.因此,深入研究金属离子对丝素蛋白构象的影响不仅可以使我们对蚕吐丝过程能有更进一步的认识,而且还可以将易于获得的稳定丝素蛋白作为模型蛋白,为中枢神经系统疾病的研究提供帮助.
