本实验采用分样筛、磁性分离和密度梯度离心综合法分离猪肾皮质的肾小管基底膜。相差显微镜观察肾小管的纯度可达97%以上。磁性分离后用超声震碎细胞核,再经密度梯度离心,即可获得高纯度的肾小管基底膜。
In this experiment, the renal tubular basement membrane of porcine renal cortex was isolated by using a sample-like sieve, magnetic separation and density gradient centrifugation. Phase contrast microscope to observe the purity of the renal tubules up to 97%. After magnetic separation, the nuclei are sonicated by ultrasound and then centrifuged by density gradient to obtain high-purity tubular basement membrane.
