目的建立同时测定大血藤各部位绿原酸、咖啡酸和香草酸含量的高效液相色谱方法。方法色谱柱为Kromasil C18柱(4.6 mm×250 mm,4.5μm),流动相:A为1%冰醋酸,B为甲醇,以A∶B=68∶32等度洗脱;柱温:30℃;流速:1 m L·min?1;检测波长:327 nm;进样量:20μL。结果在测定样品中,绿原酸和咖啡酸在乙酸乙酯部位含量最高,分别为4.52%,0.11%,香草酸在二氯甲烷部位含量最高,为5.8%。结论该方法简便、准确、重复性高,可用于大血藤中绿原酸、咖啡酸、香草酸含量的测定。
OBJECTIVE To establish a HPLC method for simultaneous determination of chlorogenic acid, caffeic acid and vanillic acid in various parts of Salvia miltiorrhiza. Methods The column was Kromasil C18 column (4.6 mm × 250 mm, 4.5 μm). The mobile phase consisted of A was 1% acetic acid and methanol was B, and was eluted with A: B = 68:32. The column temperature was 30 ℃ ; Flow rate: 1 m L · min -1; detection wavelength: 327 nm; injection volume: 20 μL. Results Chlorogenic acid and caffeic acid were the highest in the ethyl acetate fraction of the tested samples, accounting for 4.52% and 0.11% respectively. The highest content of vanillic acid in the methylene chloride was 5.8%. Conclusion The method is simple, accurate and reproducible. It can be used for the determination of chlorogenic acid, caffeic acid and vanillic acid in.
