目的探索先天性马蹄内翻足(ICTEV)与肌肉组织Fox O基因家族(Fox O1、Fox O3和Fox O4)基因表达的相关性。方法将2012~2014年新疆医科大学附属中医医院80例ICTEV患儿及40例健康儿童纳入研究,运用实时荧光定量聚合酶链反应(Real time-q PCR)和Western blot方法检测ICTEV患儿和健康儿童足部肌肉组织Fox O1、Fox O3和Fox O4的基因转录及表达水平。结果 1与健康儿童比较,ICTEV患儿肌肉组织中Fox O1基因转录水平存在明显下调,差异有统计学意义(P=0.021);Fox O3、Fox O4基因转录水平无明显变化,差异无统计学意义(P=0.738)。2ICTEV患儿肌肉组织中Fox O1蛋白表达量比健康儿童更少,差异有统计学意义(P=0.017);Fox O3、Fox O4蛋白表达水平差异无统计学意义(P=0.672)。3与汉族ICTEV患儿比较,维吾尔族ICTEV患儿肌肉组织中Fox O1基因转录水平存在显著下调;Fox O3、Fox O4基因转录水平差异无统计学意义。结论 ICTEV与Fox O基因家族之间存在相关性,Fox O1表达水平的降低可能与ICTEV的发生有关,并提示新疆地区维吾尔族ICTEV患儿更易受Fox O1表达水平的影响。
Objective To explore the relationship between congenital clubfoot (ICTEV) and muscle O Fox O gene family (Fox O1, Fox O3 and Fox O4) gene expression. Methods A total of 80 ICTEV children and 40 healthy children from the Affiliated Chinese Medicine Hospital of Xinjiang Medical University from 2012 to 2014 were enrolled in this study. Real time-q PCR and Western blot were used to detect ICTEV in children and healthy children Gene transcription and expression of Fox O1, Fox O3 and Fox O4 in foot muscles of children. Results Compared with healthy children, Fox O1 gene transcription in muscle tissue of ICTEV patients was significantly down-regulated (P = 0.021). There were no significant differences in FoxO3 and FoxO4 gene transcription (P = 0.738). The expression of FoxO1 protein in muscle tissue of 2ICTEV patients was less than healthy children, the difference was statistically significant (P = 0.017). There was no significant difference in FoxO3 and FoxO4 protein expression (P = 0.672). 3Compared with Han children with ICTEV, the transcriptional level of Fox O1 gene in muscle tissue of Uighur ICTEV children was significantly down-regulated. The transcriptional levels of Fox O3 and Fox O4 were not statistically different. Conclusions There is a correlation between ICTEV and Fox O gene family. The decrease of Fox O1 expression may be related to the occurrence of ICTEV, which suggests that the ICTEV of Uygur in Xinjiang is more likely to be affected by FoxO1 expression.
