目的研究肾移植受者的孕烷X受体(PXR)基因多态性对他克莫司浓度/剂量比的影响。方法记录患者的体重、他克莫司血药浓度等,用PCR产物直接测序法对肾移植受者进行基因分型,比较不同基因型患者间他克莫司的浓度/剂量比。结果 PXR-C24381A基因多态性与他克莫司浓度/剂量比值明显相关;他克莫司浓度/剂量比,在A/A型患者明显低于C/A型和C/C型患者(P<0.05),而PXR-A7635G基因多态性对其比值无影响(P>0.05)。结论要取得相同血药浓度,PXR-A24381A基因型患者要比PXR-C24381A型和PXR-C24381C型服用的剂量更高。
Objective To investigate the effects of polymorphism of pregnane X receptor (PXR) on the concentration / dose ratio of tacrolimus in renal transplant recipients. Methods The body weight and blood concentration of tacrolimus were recorded. Genotypes of renal transplant recipients were obtained by direct sequencing of PCR products, and the concentration / dose ratio of tacrolimus in different genotypes was compared. Results The PXR-C24381A gene polymorphism was significantly correlated with the concentration / dose ratio of tacrolimus. The concentration / dose ratio of tacrolimus was significantly lower in patients with type A / A than those with type C / C and type C <0.05), while the PXR-A7635G gene polymorphism had no effect on the ratio (P> 0.05). Conclusion To achieve the same blood concentration, patients with PXR-A24381A genotype should be given higher doses than those with PXR-C24381A and PXR-C24381C.
