目的:探讨大蒜素诱导卵巢癌细胞株OVCA-3凋亡规律。方法:应用DNA断裂点标记(TUNEL)法、DNA凝胶电泳以及光镜等技术观察有关细胞的凋亡情况。结果:0.01~100μg/ml的大蒜素作用OVCA-3细胞2h及10μg/ml的大蒜素作用OVCA-3细胞不同时间,锥虫蓝染色阳性率均在18.5%以下。TUNEL显示:1~100μg/ml的大蒜素作用OVCA-3细胞2h,即出现典型的细胞凋亡特征;50μg/ml时凋亡率为65.9%。结论:一定剂量范围内的大蒜素可在直接杀伤作用较小的情况下明显诱导OVCA-3细胞凋亡,不会因细胞坏死而引起炎症反应,这对于大蒜素应用于临床将有重要意义。
Objective: To investigate the apoptosis of OVCA-3 cells induced by allicin. Methods: Cell apoptosis was observed by DNA breakage point labeling (TUNEL), DNA gel electrophoresis and light microscopy. Results: OVCA-3 cells were treated with allicin at a dose of 0.01 ~ 100μg / ml for OVA-3 cells for 2h and 10μg / ml for allicin. The positive rates of trypan blue staining were below 18.5% at different time points. TUNEL showed that OVA-3 cells treated with allicin (1 ~ 100μg / ml) for 2h exhibited typical apoptotic characteristics. The apoptotic rate was 65.9% at 50μg / ml. CONCLUSION: Allicin in a dose range can induce apoptosis of OVCA-3 cells in a relatively small direct killing effect without causing inflammatory reaction due to cell necrosis, which is of great significance for the clinical application of allicin.
