目的:系统研究蒙古鸦葱的抗肿瘤活性成分。方法:使用硅胶、Sephadex LH-20、ODS和HPLC分离、纯化,根据理化性质和光谱分析鉴定化合物的结构。采用MTT法和SRB法进行活性跟踪和单体化合物的抗肿瘤活性筛选。结果:分离鉴定了12个化合物,分别为脱氢木香内酯(1)、β-谷甾醇(2)、胆甾醇(3)、5α,8α-环二氧-24-甲基胆甾-6,22-二烯-3β-醇(4)、硬脂酸-1-甘油单脂(5)、1-亚油酸甘油酯(6)、硬脂酸(7)、软脂酸(8)、蔗糖(9)、毛地黄黄酮-5,3′-二甲酯(10)、邻苯二甲酸二异丁酯(11)和邻苯二甲酸二正丁酯(12)。结论:1~12均为首次从蒙古鸦葱中分离得到。药理实验表明,化合物1,4和10在50μg·mL-1浓度下对小鼠肺腺癌细胞A-549显示出一定的抑制作用。
Objective: To systematically study the antitumor active ingredients of Radix Aconitum Methods: Silica gel, Sephadex LH-20, ODS and HPLC were used to separate and purify the compounds. The structures of the compounds were identified by their physicochemical properties and spectral analysis. MTT assay and SRB method were used to determine the activity and the antitumor activity of monomeric compounds. RESULTS: Twelve compounds were isolated and identified as dehydrocostus (1), β-sitosterol (2), cholesterol (3), 5α, 8α-cyclooxygen-24-methylcholesterol- Glyceryl monostearate (5), glycerol 1-linoleate (6), stearic acid (7), palmitic acid ), Sucrose (9), luteolin-5,3’-dimethyl ester (10), diisobutyl phthalate (11) and di-n-butyl phthalate (12). Conclusion: 1 ~ 12 are all isolated from Carambola for the first time. Pharmacological experiments showed that compounds 1, 4 and 10 showed a certain inhibitory effect on mouse lung adenocarcinoma A-549 cells at the concentration of 50μg · mL-1.
