目的观察外源性3-脱氧葡萄糖醛酮(3-DG)对正常小鼠血糖的影响。方法雄性昆明种小鼠12只,随机分为3-DG组(5mg/kg)和对照组(Con组),各6只。灌胃3-DG第1天、1周、2周后分别采用血糖仪测定小鼠自由饮食和空腹状态下的血糖;并对灌胃3-DG2周的小鼠进行糖耐量实验。结果(1)小鼠单次灌服3-DG后2h的血糖比灌胃前升高(7.78±0.694 vs 6.41±0.408,P<0.05),与Con组比显著升高(7.78±0.694vs 6.43±0.427,P<0.01);连续灌服3-DG 1周,3-DG组多时间点的血糖均比Con组升高,但差异无统计学意义(P>0.05)。(2)小鼠单次灌服3-DG后空腹6h与连续灌服3-DG 2周后空腹血糖与Con组比,均无统计学差异(P>0.05)。(3)小鼠连续灌服3-DG 2周,与Con组比,糖耐量30min(19.47±1.541 vs 17.22±1.911,P<0.05)、60min血糖(P>0.05)升高,120min血糖无变化(P>0.05)。结论外源性3-DG能够升高正常小鼠血糖水平,主要表现在单次灌胃3-DG对小鼠血糖有即时升高的作用和连续灌胃引起血糖生理曲线上移及糖耐量轻度受损。
Objective To observe the effect of exogenous 3-deoxyglucosone (3-DG) on blood glucose in normal mice. Methods Twelve male Kunming mice were randomly divided into 3-DG group (5mg / kg) and control group (Con group), 6 mice in each group. Gavage 3-DG on day 1, week 1, and week 2 were used to measure blood glucose in free diet and fasting state, respectively. Glucose tolerance test was performed in mice given 3-DG for 3 weeks. Results (1) Compared with the Con group, the blood glucose level of mice after 3-DG administration was significantly higher than that before Constipation (7.78 ± 0.694 vs 6.41 ± 0.408, P <0.05) ± 0.427, P <0.01). Compared with Con group, the blood glucose in 3-DG group was significantly increased at 3-DG administration for 3 weeks (P> 0.05). (2) There was no significant difference (P> 0.05) between fasting blood glucose and Con group after 3-DG fasting 6h and continuous 3-DG 2 weeks. (3) The mice were continuously fed with 3-DG for 2 weeks. Compared with the Con group, the mice were given glucose tolerance for 30min (19.47 ± 1.541 vs 17.22 ± 1.911, P <0.05) (P> 0.05). Conclusion Exogenous 3-DG can raise the blood glucose level of normal mice, which is mainly manifested as the instantaneous increase of blood glucose in a single oral administration of 3-DG and the up-regulation of blood glucose physiological function and glucose tolerance Degree of damage.
