目的阐明大承气汤经灌胃给药后,5种游离蒽醌成分在大鼠体内的药物动力学特性。方法大鼠灌胃大承气汤9 g/kg后,采集血浆,采用HPLC法测定血浆中芦荟大黄素、大黄酸、大黄素、大黄酚及大黄素甲醚的含量,并用3p87软件进行数据分析。结果大鼠灌胃大承气汤后,血浆中芦荟大黄素、大黄酸、大黄素、大黄酚及大黄素甲醚的T1/2(Ka)依次是0.362,1.034,1.918,1.89,0.656 h;T1/(2Ke)依次是0.311,1.173,2.33,2.188,0.81 h;Cmax依次是48.47,21.69,10.49,5.76,38.76 mg/L;Tmax依次是4,4,6,4,2 h;AUC0-t依次是85.733,66.726,65.911,41.84,96.401 mg.L-1.h-1。结论通过建立经灌胃大承气汤后大鼠血浆中5种游离蒽醌成分含量的测定方法,阐明其药动学特性,为经典寒下方大承气汤的体内成分研究提供实验依据。
Objective To elucidate the pharmacokinetics of 5 kinds of free anthraquinones in rats after oral administration of Dachengqi Decoction. Methods After the rats were inoculated with Dachengqi Decoction at a dose of 9 g / kg, the plasma was collected and the contents of aloe-emodin, rhein, emodin, chrysophanol and physcion in plasma were determined by HPLC. The data were analyzed by 3p87 software . Results T1 / 2 (Ka) of aloe-emodin, rhein, emodin, chrysophanol and physcion in plasma were 0.362,1.034,1.918,1.89 and 0.656 h after rats were administered with Dachengqi Decoction. T1 / (2Ke) were 0.311,1.173,2.33,2.188,0.81 h; Cmax followed by 48.47,21.69,10.49,5.76,38.76 mg / L; Tmax followed by 4,4,6,4,2 h; AUC0- t followed by 85.733, 66.726, 65.911, 41.84, 96.401 mg.L-1.h-1. Conclusion The method of determining the content of five free anthraquinones in the plasma of rats through the method of intragastric administration of Dachengqi Decoction is established to clarify the pharmacokinetic characteristics and provide the experimental evidence for the in vivo study of the classic Dwarfing Decoction.
