目的建立超声辅助提取焙干鹿茸中次黄嘌呤的方法。方法采用正交实验设计系统研究了试剂浓度、试剂体积、超声时间、超声温度对鹿茸中次黄嘌呤的提取产生的影响。结果 1.000g焙干鹿茸样品用40ml50%乙醇60℃提取40min时,提取液中次黄嘌呤含量最多。超声辅助提取鹿茸中的次黄嘌呤,采用反相高效液相色谱-紫外检测器进行测定,在1~30mg/L浓度范围内,其峰面积与浓度具有良好的线性关系,加标回收率97.5%~102.9%。结论超生辅助提取次黄嘌呤,反相高效液相色谱法测定可以快速、准确测定经焙干处理后鹿茸样品中次黄嘌呤的含量。
Objective To establish a sonication-assisted extraction of hypoxanthine in pilose antler. Methods The effects of reagent concentration, reagent volume, ultrasonic time and ultrasonic temperature on the extraction of hypoxanthine in pilose antler were systematically studied by orthogonal experimental design. Results 1.000g dried antler samples with 40ml50% ethanol at 60 ℃ for 40min, the extract most hypoxanthine content. Hypoxanthine in antler was extracted by ultrasound, and the linearity was determined by reversed-phase high performance liquid chromatography-ultraviolet detector. The peak area and concentration were linear in the range of 1 ~ 30 mg / L with the recoveries of 97.5 % ~ 102.9%. Conclusions Auxiliary extraction of hypoxanthine by reversed-phase high performance liquid chromatography can quickly and accurately determine the content of hypoxanthine in the antler samples after drying.
