目的探讨乳腺癌患者白细胞介素6(IL-6)水平及IL-6基因启动区-597 G/A、-572 C/G、-174 G/C三个单核苷酸多态性(SNP)与乳腺癌的相关性。方法用DNA测序法检测136例乳腺癌患者及150例无血缘关系正常对照组IL-6基因SNP位点的基因多态性,电化学发光法检测血清IL-6水平。应用χ2检验法分析各位点基因型分布与乳腺癌发病风险的相关性,并探讨其与临床病理特征的关系。各组间IL-6水平的差异分别采用t检验法进行统计学分析。结果乳腺癌组IL-6水平显著高于正常对照组,IL-6基因-572 C/G基因型乳腺癌患者中的频率显著高于对照人群(OR=1.841,95%CI:1.115~3.040,P=0.017)。乳腺癌组及正常对照组-572 C/G位点不同基因型之间的IL-6水平均无显著性差异。结论乳腺癌患者IL-6水平显著升高,提示IL-6表达水平及-572 C/G基因多态性在乳腺癌的发病中起一定作用。
Objective To investigate the levels of interleukin 6 (IL-6) and three SNPs (-597 G / A, -572 C / G and -174 G / C) in promoter region of IL- ) And breast cancer. Methods The gene polymorphism of SNP in IL-6 gene was detected by DNA sequencing in 136 cases of breast cancer and 150 unrelated normal controls. The level of IL-6 in serum was detected by electrochemiluminescence. Chi-square test was used to analyze the relationship between genotype distribution and the risk of breast cancer, and to explore its relationship with clinicopathological features. The differences of IL-6 levels between groups were analyzed by t-test. Results The level of IL-6 in breast cancer group was significantly higher than that in control group. The frequency of IL-6 gene -572 C / G breast cancer was significantly higher than that in control group (OR = 1.841, 95% CI: 1.115-3.040, P = 0.017). There was no significant difference in IL-6 levels between different genotypes at -572 C / G in breast cancer group and normal control group. Conclusion The level of IL-6 in patients with breast cancer is significantly increased, suggesting that the level of IL-6 and the -572 C / G polymorphism play a role in the pathogenesis of breast cancer.
