[目的]分离纯化北五味子茎藤中具有抗肝癌细胞活性的化学成分.[方法]反复采用硅胶柱色谱及重结晶等方法分离化合物.根据理化性质和波谱学分析,鉴定化学结构.并采用MTT法分析化合物的体外抗肝癌细胞作用.[结果]分离得到7个化合物,分别鉴定为五味子丙素(Ⅰ)、十九醇(Ⅱ)、三十五醇(Ⅲ)、二十四醇(Ⅳ)、五味子乙素(Ⅴ)、β-谷甾醇(Ⅵ)及五味子醇乙(Ⅶ).活性检测结果见,化合物Ⅰ,Ⅴ,Ⅵ有较强的体外抗肝癌细胞活性,质量浓度均为50mg/L时的抑制率分别为96.27%,97.11%,93.99%;化合物Ⅱ,Ⅲ,Ⅳ,Ⅶ的质量浓度均为50mg/L时抑制率分别为86.81%,82.33%,77.62%,86.25%.[结论]从北五味子茎藤中分离得到的7个化合物均具有体外抗肝癌细胞活性.
[Objective] The research aimed to isolate and purify the chemical constituents of Hedyotis sibiricus with anti-hepatocarcinoma cell activity. [Method] The compounds were separated and purified repeatedly by silica gel column chromatography and recrystallization. The chemical structures were identified by physicochemical and spectroscopic analysis. MTT [Methods] Seven compounds were isolated and identified as Schisandra promelic (Ⅰ), nonadecyl (Ⅱ), pentadecyl alcohol (Ⅲ), and tetracosanol Ⅳ ), Schisandrin B (Ⅴ), β-Sitosterol (Ⅵ) and Schisandra chinensis B (Ⅶ) .The results of activity test showed that the compounds Ⅰ, Ⅴ and Ⅵ had strong anti-hepatocarcinoma cell activity in vitro with mass concentration of 50mg / L, the inhibitory rates were 96.27%, 97.11%, 93.99%, respectively. The inhibitory rates were 86.81%, 82.33%, 77.62% and 86.25% when the concentration of compound Ⅱ, Ⅲ, Ⅳ and Ⅶ were both 50mg / L. [Conclusion] The seven compounds isolated from the stems of Schisandra chinensis possess anti-hepatocellular carcinoma cell activity in vitro.
