目的:解决野生药用植物黄花石蒜外植体大小影响其组织培养效果的难题。方法:选用其鳞茎的带底盘双内层鳞片、带底盘双中层鳞片为材料,采用长宽为9.0×8.0mm、6.0×5.0mm、3.0×2.0mm3种不同大小的外植体,接种在MS+9mg/LBA+5mg/LNAA+0.7%琼脂+3%蔗糖与MS+5mg/LBA+9mg/LNAA+0.7%琼脂+3%蔗糖2种培养基上进行培养,研究了黄花石蒜外植体的不同大小对其组织培养效果的影响。结果与结论:6.0×5.0mm中等大小的带底盘双内层鳞片外植体在MS+9mg/LBA+5mg/LNAA+0.7%琼脂+3%蔗糖的培养基上培养有利于不定芽、不定根的发生,产生褐变量也较少。相关分析表明,不同大小的2种外植体在2种培养基上的培养效果为极显著正相关,这进一步说明,在一种培养基中培养效果较好的外植体大小在另一种培养基中其培养效果仍然较好。6.0×5.0mm中等大小的带底盘双内层鳞片外植体的培养效果较其他外植体好,其次为6.0×5.0mm中等大小的带底盘双中层鳞片外植体。
OBJECTIVE: To solve the problem that the size of wild Lycoris radiata explants on tissue culture effect.
