目的:建立RP-HPLC法测定盐酸阿霉素磁性脂质体的药物含量。方法:采用Agilent HC-C18柱(250mm×4.6mm,5μm);流动相:甲醇-醋酸钠缓冲液(冰醋酸调pH3.4)(7:3);流速:1.0mL.min-1;紫外检测波长:233nm;柱温:室温。结果:盐酸阿霉素浓度在2.0~50.0μg.mL-1范围内线性关系良好(r=0.9994,n=7),平均回收率为97.1%。结论:本方法简便快速、精密可靠,可用于盐酸阿霉素磁性脂质体中盐酸阿霉素含量的测定。
Objective: To establish a RP-HPLC method for the determination of doxorubicin hydrochloride magnetic liposomes. The mobile phase consisted of methanol-sodium acetate buffer (adjusted to pH3.4 with glacial acetic acid) (7: 3); flow rate: 1.0 mL · min-1; UV Detection wavelength: 233nm; column temperature: room temperature. Results: The linearity of doxorubicin hydrochloride in the range of 2.0-50.0μg.mL-1 was good (r = 0.9994, n = 7). The average recovery was 97.1%. Conclusion: The method is simple, rapid, accurate and reliable and can be used for the determination of doxorubicin hydrochloride in doxorubicin hydrochloride magnetic liposomes.
