目的:观察不同浓度的复方半枝莲对肝纤维化大鼠模型肝脏组织及基质金属蛋白酶-2(MMP-2)的影响,探讨复方半枝莲防治肝纤维化的效果。方法:采用皮下注射40%CCl4配合高脂饲料和酒精干预方法建立大鼠肝纤维模型,自造模之日起各组每天早上分别空腹灌胃生理盐水、复方半枝莲低剂量溶液、复方半枝莲中剂量溶液、复方半枝莲高剂量溶液、秋水仙碱混悬液,共灌胃给药4 w后,光镜观察肝组织病理变化;放射性免疫方法(RIA)检测PCⅢ、ColⅣ、HA、LN的含量;免疫组织化学方法(IHC)检测肝组织MMP-2的表达,以半定量方法评定。结果:经复方半枝莲干预后,复方半枝莲高剂量组中MMP-2含量明显减少,与秋水仙碱组比较有显著性(P<0.01)。结论:复方半枝莲能够改善肝纤维化大鼠肝功能,降低其血清肝纤维化指标PCⅢ、ColⅣ、HA、LN水平,有效改善肝纤维化大鼠肝组织病理学损害,与复方半枝莲能降低MMP-2的表达有关。
Objective: To observe the effects of different concentrations of Scutellaria barbata on liver tissue and matrix metalloproteinase-2 (MMP-2) in rats with hepatic fibrosis and to explore the effect of Scutellaria barbata against hepatic fibrosis. Methods: The rat model of hepatic fibrosis was established by subcutaneous injection of 40% CCl4 with high-fat diet and alcohol intervention. From the day of modeling, rats in each group were given fasting gavage with normal saline, compound Scutellaria barbata low dose solution, compound half The middle dose of Scutellaria barbata, the high dose of compound Scutellaria barbata and the colchicine suspension for 4 w, the histopathological changes of liver were observed under light microscope. Radioimmunoassay (RIA) was used to detect the expression of PCⅢ, Col Ⅳ, HA , And LN. The expression of MMP-2 in liver tissues was detected by immunohistochemical method (IHC) and evaluated by semi-quantitative method. Results: After the intervention of compound Scutellaria barbata, the content of MMP-2 in compound Scutellaria barbata high dose group obviously decreased, compared with the colchicine group (P <0.01). Conclusion: Scutellaria barbata can improve hepatic function, reduce the level of PCⅢ, ColⅣ, HA and LN in liver fibrosis rats and effectively improve the pathological damage of hepatic tissue in rats with hepatic fibrosis. Can reduce the expression of MMP-2.
