目的探讨在不同正畸力作用下龈沟液内白介素-17的质量浓度变化,建立大鼠正畸牙根吸收模型。方法选择体质量相近、月龄相同的SD雄性成年大鼠建立正畸牙根吸收模型,按不同正畸作用力分为对照组、40g组、60g组及80g组4个组。加力14 d后,采用连续切片行苏木素-伊红染色观察牙根吸收情况,提取实验牙龈沟液,应用免疫组织化学染色方法及双抗夹心酶联免疫吸附法(ELISA)观察白介素-17的表达变化情况。结果牙根吸收主要表达在根中1/3区域且吸收程度与力值有关;白介素-17的表达与力的大小之间具有相关性(P<0.05),有统计学意义,且与力的大小呈正相关;根吸收相对面积与白介素-17阳性表达率呈正相关(P<0.05)。结论白介素-17可能参与了正畸牙根吸收过程。白介素-17参与正畸牙移动过程中牙周组织改建,提示临床操作中应注意加力大小控制。
Objective To investigate the changes of the concentration of interleukin-17 in gingival crevicular fluid under different orthodontic forces, and establish the model of orthodontic root resorption in rats. Methods SD adult male SD rats with similar body mass and age were selected to establish orthodontic tooth root resorption model and divided into 4 groups according to different orthodontic forces: control group, 40g group, 60g group and 80g group. After 14 days of force application, the root resorption was observed by hematoxylin-eosin staining and the experimental gingival crevicular fluid was extracted. The expression of interleukin-17 was detected by immunohistochemical staining and double antibody sandwich enzyme-linked immunosorbent assay (ELISA) Changes. Results The root resorption was mainly expressed in 1/3 of the root area and the degree of absorption was related to the force value. There was a significant correlation between the expression of interleukin-17 and the size of the force (P <0.05) (P <0.05). There was a positive correlation between the relative area of root resorption and the positive rate of interleukin-17 (P <0.05). Conclusion IL-17 may be involved in the orthodontic root resorption process. Interleukin-17 is involved in periodontal tissue remodeling during orthodontic tooth movement, suggesting that the force control should be emphasized in clinical practice.
