目的:探讨针刺治疗CFS模式大鼠对其海马与下丘脑中Smad4蛋白表达的作用及其机制.方法:清洁级SD雄性大鼠48只,随机分为正常组、模型组、束缚组、针刺组,采用游泳多重应激刺激建立慢性疲劳综合征动物模型,针刺组选择百会、情感区、感觉区、给予电针治疗,采用Western blot技术检测各组Smad4蛋白表达水平;RT-PCR技术检测各组Smad4 mRNA表达水平.结果:western blot检测显示:CFS模型组大鼠海马与下丘脑中Smad4蛋白表达明显增多,而正常组则表达较低,针刺治疗后,针刺组大鼠海马与下丘脑中Smad4蛋白表达明显降低;RT-PCR法检测大显示:各组大鼠海马和下丘脑中均有Smad4mRNA表达,正常组Smad4 mRNA表达较弱,模型组Smad4 mRNA表达明显增加,治疗后,针刺组的Smad4mRNA表达水平明显降低.结论:电针百会、情感1区和感觉区能够降低CFS模型大鼠海马与下丘脑中Smad4 mRNA水平,从而抑制Smad4蛋白表达,进而对CFS疲劳状态起到改善和治疗作用.
