本文对新疆境内307例维吾尔族无关个体D6S1043、PentaE两个STR基因座遗传多态性进行调查,为相关研究与应用提供参考数据。1材料与方法 1.1样本及DNA提取307例无关个体血样全部来自于日常检案中犯罪嫌疑人和受害人,采静脉血制成血痕保存。采用Chelex-100法提取样本DNA。1.2 PCR扩增及检测应用DNATyperTM15试剂盒,扩增体系为10μL,内含PCR Reaction Mix 2.0μL,Primer 1.0μL,Taq酶0.2μL,去离子水6μL,模板DNA 0.8μL。PCR反应在9700型扩增仪(美国AB公司)中进行,循环条件试剂盒默认条件。
In this paper, we investigated the genetic polymorphism of two STR loci in 307 Uighur unrelated individuals in Xinjiang Uygur Autonomous Region, and provided reference data for related research and application. 1 Materials and Methods 1.1 Samples and DNA Extraction 307 unrelated individual blood samples all from the daily prosecution of suspects and victims, collecting venous blood made of blood stains. The sample DNA was extracted using the Chelex-100 method. 1.2 PCR amplification and detection DNATyperTM15 kit, the amplification system was 10μL containing PCR Reaction Mix 2.0μL, Primer 1.0μL, Taq enzyme 0.2μL, deionized water 6μL, template DNA 0.8μL. The PCR reaction was performed in a Model 9700 Thermal Cycler (AB USA) with the default conditions of the cycling kit.
