目的探讨IL-10区域动脉灌注(LAI)对急性胰腺炎(AP)大鼠血浆IL-6和TNF-α的影响。方法以5%牛磺胆酸钠胰胆管注射(1 ml/kg)制成大鼠急性胰腺炎模型,IL-10(40 000 UI/kg)LAI,以生理盐水为对照,检测大鼠血浆IL-10I、L-6、TNF-α和淀粉酶。结果假手术组大鼠血浆IL-10测不出,IL-6、TNF-α和淀粉酶分别为(17.5±1.3)pg/ml、(71.25±5.75)pg/ml和(885.4±136.3)U/L,AP大鼠血浆IL-6、TNF-α明显升高,与假手术组对比有显著差异(P<0.05);IL-10 LAI则使AP大鼠血浆3 h和6h的IL-6、TNF-α明显降低,与生理盐水组及AP组对比均有显著差异(P<0.05),但12 h时二者明显升高,与生理盐水组及AP组对比均有显著差异(P<0.05),IL-10LAI使大鼠淀粉酶明显下降。结论IL-10 LAI能降低AP大鼠3 h和6 h时的IL-6、TNF-α,但后期(12 h)则使二者升高,同时能降低AP大鼠淀粉酶,IL-10有望成为AP综合治疗的药物之一。
Objective To investigate the effect of IL-10 regional arterial infusion (LAI) on plasma IL-6 and TNF-α in rats with acute pancreatitis (AP). Methods Rat acute pancreatitis model was induced by 5% sodium taurocholate (1 ml / kg) injection. IL-10 (40 000 UI / kg) LAI was injected into the rats. -10I, L-6, TNF-α and amylase. Results The levels of IL-6, TNF-α and amylase in the sham operation group were (17.5 ± 1.3) pg / ml, (71.25 ± 5.75) pg / ml and (885.4 ± 136.3) U (P <0.05). IL-10 and LAI induced a significant increase of plasma IL-6 and TNF-α in AP rats at 3 h and 6 h (P <0.05), but both of them were significantly increased at 12 h (P <0.05), compared with the normal saline group and the AP group (P < 0.05), IL-10LAI significantly decreased amylase in rats. Conclusion IL-10 LAI can reduce IL-6 and TNF-α at 3 h and 6 h in AP rats, but increase IL-6 LAI at 12 h in AP rats, meanwhile decrease amylase, IL-10 It is expected to become one of the comprehensive treatment of AP drugs.
