目的探讨蛋白酪氨酸磷酸酶1B(PTP1B)在乳腺非特殊型浸润性癌(NST)组织中的表达及意义。方法采用原位杂交技术和免疫组化法分别检测86例乳腺NST组织中PTP1B mRNA和蛋白的表达,并分析其与乳腺癌临床病理特征的关系,以癌旁正常乳腺组织作为阴性对照。结果 PTP1B mRNA及蛋白在乳腺癌组织中的阳性表达率分别为52.3%和59.3%,均明显高于癌旁正常乳腺组织,差异有统计学意义(P≤0.01)。PTP1B mRNA与蛋白表达呈正相关(rs=0.773,P<0.001),且PTP1B表达与乳腺癌淋巴结转移、TNM分期有关(P≤0.001),与者患者年龄、肿瘤大小和组织学分级无关(P均>0.05)。生存分析结果显示,PTP1B蛋白阳性患者的生存时间短于阴性患者(χ~2=13.071,P<0.001)。结论 PTP1B在乳腺癌组织中高表达,且与淋巴结转移、TNM分期有关,其有望成为乳腺癌治疗的潜在靶点。
Objective To investigate the expression and significance of protein tyrosine phosphatase 1B (PTP1B) in breast non-special invasive carcinoma (NST). Methods The expression of PTP1B mRNA and protein in 86 cases of breast NST tissues were detected by in situ hybridization and immunohistochemistry respectively. The relationship between the expression of PTP1B mRNA and the clinicopathological features of breast cancer was analyzed. The normal breast tissue adjacent to cancer was taken as the negative control. Results The positive rates of PTP1B mRNA and protein in breast cancer tissues were 52.3% and 59.3%, respectively, which were significantly higher than those in normal breast tissues (P≤0.01). PTP1B mRNA was positively correlated with protein expression (rs = 0.773, P <0.001). PTP1B expression was correlated with lymph node metastasis and TNM stage in breast cancer (P≤0.001), but not with age, tumor size and histological grade > 0.05). Survival analysis showed that the survival time of PTP1B-positive patients was shorter than that of negative patients (χ ~ 2 = 13.071, P <0.001). Conclusions PTP1B is highly expressed in breast cancer tissues and is related to lymph node metastasis and TNM stage. It is expected to be a potential target for the treatment of breast cancer.
