本实验采用PI与ANNEXIN-v双染法,用流式细胞仪检测益气通络丹合药血清对缺氧-再给氧心肌 细胞凋亡的作用。实验结果表明:缺氧24h再给氧4h能导致体外培养心肌细胞凋亡,提取DNA电泳表现为梯 形特征(LADDER);益气通络丹含药血清能显著降低缺氧-再给氧心肌细胞的凋七比率,流式细胞仪检测结果 表明,益气通络丹低、中、高剂量含药血清细胞凋亡率较缺氧-再给氧组明显下降(P<0.01),其作用与剂量相 关。提示益气通络丹抗心肌缺血再灌注损伤作用与抑制心肌细胞凋亡有关,可能是该方延缓缺血性心脏病渐 进性病理损害的重要机制之一。
In this experiment, PI and ANNEX IN-v double staining method was used to detect the effect of serum of Yiqi Tongluo Tang mixture on the apoptosis of myocardial cells induced by hypoxia-reoxygenation by flow cytometry. The experimental results showed that the cardiomyocytes apoptosis in vitro was induced by hypoxia for 24h and then oxygen for 4h. The DNA electrophoresis showed a trapezoidal feature (LADDER); Yiqi Tongluo Dan serum could significantly reduce the hypoxia-reoxygenated cardiomyocytes. The withered ratio, flow cytometry results showed that the apoptosis rate of low-, medium-, and high-dose drug-containing serum in Yiqi Tongluo Dan was significantly lower than that in the hypoxia-reoxygenation group (P<0.01). The effect is related to the dose. It is suggested that the effect of Yiqi Tongluodan against myocardial ischemia-reperfusion injury is related to the inhibition of myocardial cell apoptosis, which may be one of the important mechanisms for delaying the progressive pathological damage of ischemic heart disease.
