目的探讨中药促愈熏洗方对大鼠背部早期创面愈合的影响,并通过实验研究其促进愈合的机制。方法选取Wistar大鼠制成全层皮肤缺损模型,随机分为中药组(促愈熏洗方)6只、西药组(利凡诺)6只、空白对照组(0.9%Na Cl溶液)6只,进行相应干预。分别观察各组创面愈合情况,并用Western blotting法检测创面组织病理改变VEGF、VEGFR-1、VEGFR-2及血清酶联免疫法(ELISA)检测TNF-α、IL-6的表达变化。结果中药组创面愈合时间与西药组基本相同,均短于空白对照组,造模后第3 d各观察指标无统计学意义(P>0.05);第7、15 d促愈熏洗方组创面愈合率及肉芽生长情况、血清TNF-α、IL-6水平均优于空白对照组(P<0.05)。第7 d,中药组创面愈合率高于西药组(P<0.05)。各组VEGF、VEGFR-1、VEGFR-2表达差异均无统计学意义(P>0.05)。结论促愈熏洗方可明显抑制创面早期炎症因子TFN-α和IL-6的表达,从而促进创面的愈合。
Objective To investigate the effect of “Yuxu Fu-Fang Decoction” on wound healing in rats with early wound healing and to study its mechanism of promoting healing through experiments. Methods Wistar rats were selected to make a full-thickness skin defect model. They were randomly divided into six groups: Chinese herbal medicine group (Yuxufuxu recipe), western medicine group (Rivanol) and blank control group (0.9% Na Cl solution) , The corresponding intervention. The wound healing of each group was observed. The expressions of VEGF, VEGFR-1, VEGFR-2 and TNF-α and IL-6 in wound tissues were detected by Western blotting. Results The wound healing time of the traditional Chinese medicine group was basically the same as that of the western medicine group, both of which were shorter than that of the blank control group. There was no significant difference on the 3rd day after the model was established (P> 0.05) The healing rate and granulation growth, serum TNF-α, IL-6 levels were better than the blank control group (P <0.05). On the 7th day, the wound healing rate in the traditional Chinese medicine group was higher than that in the Western medicine group (P <0.05). The expression of VEGF, VEGFR-1 and VEGFR-2 in each group had no statistical significance (P> 0.05). Conclusion Yuxufang can significantly inhibit the expression of TFN-α and IL-6, and promote the wound healing.
