目的探讨触珠蛋白(Haptoglobin,Hp)在急性心肌梗死(Acute myocardial infarction,AMI)患者血清中的表达情况,为该疾病的诊断与治疗提供新思路。方法收集30例确诊的AMI患者和30例体检证实为健康人群组的血清,去除血清高丰度蛋白后,采用双向电泳(2-DE)分离血清蛋白,获得的电泳图谱经ImageMaster5.00软件分析,差异明显的蛋白点经胶内酶解后,行基质辅助激光解吸电离飞行时间质谱(Matrix-assisted laser desorption-ionization-time of flight-mass spectrometry,MALDI-TOF-MS)分析,检测Hp的表达。结果 AMI患者与健康人群组血清的电泳图谱相似;MALDI-TOF-MS分析显示,AMI患者血清中Hp表达量明显高于健康人群组(P<0.05)。结论 Hp在AMI患者血清中表达量升高,其有望成为AMI患者的特异性诊断标记物。
Objective To investigate the expression of haptoglobin (Hp) in the serum of patients with acute myocardial infarction (AMI) and to provide new ideas for the diagnosis and treatment of the disease. Methods Serum samples from 30 patients with AMI and 30 healthy people were collected. Serum protein was separated by two-dimensional electrophoresis (2-DE) after removal of serum high abundance protein. The electrophoretogram obtained by ImageMaster5.00 software The protein spots with obvious difference were analyzed by matrix-assisted laser desorption-ionization-time of flight-mass spectrometry (MALDI-TOF-MS) expression. Results The serum electrophoresis patterns of AMI patients were similar to those of healthy people. MALDI-TOF-MS analysis showed that the expression of Hp in AMI patients was significantly higher than that in healthy people (P <0.05). Conclusion The expression of Hp in the serum of patients with AMI is elevated, which is expected to become a specific diagnostic marker in AMI patients.
