本实验采用剂量递增法 ,海洛因的给药剂量从 1 mg/ kg递增至 8mg/ kg,维持8mg/ kg到用药 42天后建立的大鼠依赖模型。经注射 (i p)纳洛酮 2 mg/ Kg催促后 ,雌鼠的主要戒断症状为扭体反应 ,并具有明显的激惹症状 ,较少出现腹泻症状 ,体重变化不明显 ;而雄鼠主要表现为腹泻症状和体重下降。实验观察和实验数据均表明 ,本实验所建立的大鼠海洛因依赖模型是可靠的 ,但应注意大鼠性别上的差异可以导致海洛因依赖大鼠戒断得分的显著性差异。由此认为 ,进行大鼠海洛因催促戒断实验时 ,实验鼠应雌雄各半 ,如有条件全部采用雄性大鼠亦可。
In this study, the dose escalation method was used. The dose of heroin was increased from 1 mg / kg to 8 mg / kg, and the rat-dependent model was established after 8 days. Experimental observations and experimental data indicate that the heroin dependence model established in this experiment is reliable, but it should be noted that differences in the sex of rats can lead to significant differences in heroin dependence rats withdrawal score. Therefore, for rats heroin urged abstinence experiment, the experimental mice should be male and female, if conditions can be all male rats.
