目的:观察平滑肌中电导钙激活钾通道(KCa3.1)在糖尿病血管并发症发生中的作用与机制。方法:高糖高脂饲料喂养继STZ注射制备糖尿病大鼠模型,成模12周后检测血生化指标,行胸主动脉HE、弹力纤维、Masson染色和平滑肌α-actin、KCa3.1免疫组化染色,Western blotting检测主动脉中层和糖基化终产物(AGEs)孵育的血管平滑肌细胞(VSMCs)KCa3.1表
Objective: To observe the effect and mechanism of calcium-activated potassium channel (KCa3.1) in the development of diabetic vascular complications in smooth muscle. Methods: The diabetic rats were fed with high glucose and high fat diet following STZ injection. Blood biochemical parameters were measured 12 weeks after model injection. Thoracic aorta HE, elastic fibers, Masson staining and smooth muscle α-actin, KCa3.1 immunohistochemistry Staining and Western blotting were used to detect the expression of KCa3.1 in vascular smooth muscle cells (VSMCs) incubated with middle aorta and glycosylation end products (AGEs)
