目的:探讨中药复方益糖康对3T3-L1前脂肪细胞增殖和分化的影响。方法:培养3T3-L1前脂肪细胞;以四甲基偶氮唑盐(MTT)的方法检测益糖康对3T3-L1前脂肪细胞增殖的影响,通过流式细胞仪检测细胞增殖的周期;以油红O染色异丙醇萃取的方法检测益糖康对3T3-L1前脂肪细胞分化过程中脂肪积聚的影响。结果:10%含益糖康的大鼠血清,对3T3-L1前脂肪细胞的增殖影响最佳,细胞增殖在S、G2期与对照组比较增多;5%含益糖康大鼠血清抑制分化过程中的脂肪积聚最佳。结论:中药复方益糖康具有促进3T3-L1前脂肪细胞增殖,抑制3T3-L1前脂肪细胞分化的作用。
OBJECTIVE: To investigate the effect of traditional Chinese medicine compound Yikangkang on the proliferation and differentiation of 3T3-L1 preadipocytes. Methods: 3T3-L1 preadipocytes were cultured; the effect of Yikangkang on the proliferation of 3T3-L1 preadipocytes was detected by MTT assay, and the cycle of cell proliferation was detected by flow cytometry; The effect of Yikangkang on the fat accumulation in the differentiation of 3T3-L1 preadipocytes was detected by oil red O staining with isopropyl alcohol. Results: The serum containing 10% Yikangkang had the best effect on the proliferation of 3T3-L1 preadipocytes. The cell proliferation was increased in S and G2 phases compared with the control group. The serum of 5% Yikangkang rats was inhibited from differentiating. The fat accumulation during the process is optimal. Conclusion: Yifukang, a traditional Chinese medicine compound, can promote the proliferation of 3T3-L1 preadipocytes and inhibit the differentiation of 3T3-L1 preadipocytes.
