本研究以5个春麦品种的幼穗为外植体,接种在MS为基本培养基不同浓度的激素的诱导培养基上,结果发现,铁春2号是最好的基因型,诱导率、分化率最高,不同品种间诱导率存在极显著差异,6-BA不利于愈伤组织诱导,最佳生长素配比是20mgL-12,4-D+05mgL-1NAA。在分化培养中,最好的激素配比为02mgL-1NAA+01mgL-1Kt。经长期继代的愈伤组织5℃低温处理10d,显著提高愈伤组织的分化率。
In this study, young ears of five spring wheat varieties were used as explants and inoculated on the induction medium of different concentrations of hormones with MS as the basic medium. It was found that Tiechun 2 was the best genotype, induction rate, differentiation 6-BA is not conducive to callus induction, the best auxin ratio is 2 0 mg L-12, 4-D + 0 5 mg L-1 NAA. In differentiation culture, the best hormone ratio was 0 2 mg L-1 NAA +0 1 mg L-1 Kt. The long-term subculture of callus 5 ℃ low temperature treatment 10d, significantly increased the callus differentiation rate.
