目的观察HIV/HCV重叠感染和高效抗反转录病毒治疗(HAART)对HCV准种的影响。方法通过PCR、测序及单链构象多态性分析建立HCV高变1区(HVR1)准种变异率检测方法,运用该方法对我国上海地区48例HIV/HCV重叠感染者HCV准种变异的分子流行病学进行研究。结果与单独HCV感染组和HIV/HCV重叠感染且已行HAART组相比较,HIV/HCV重叠感染但未行HAART组的HCV准种变异率较低,差异有统计学意义(P均<0.05)。单独HCV感染组与HIV/HCV重叠感染且已行HAART组之间作比较,HCV准种的复杂性和多样性差异均无统计学意义。结论 HIV感染可降低HCV准种变异率。随着HAART长期应用,HIV/HCV重叠感染者的HCV准种率增加。
Objective To investigate the effects of HIV / HCV infection and highly active antiretroviral therapy (HAART) on HCV quasispecies. Methods The mutation rate of HVR1 in HVR1 was detected by PCR, sequencing and single-strand conformation polymorphism analysis. The molecular variation of HCV in 48 HCV / HCV co-infected patients in Shanghai area of China Epidemiology study. Results Compared with HAART group and HCV infection group and HIV / HCV overlap group, the variation rate of HCV quasispecies was lower in HIV / HCV infected but not in HAART group (P <0.05) . There was no significant difference in the complexity and diversity of HCV quasispecies between HCV infection alone and HIV / HCV overlap infection and between HAART groups. Conclusion HIV infection can reduce the variability of quasispecies of HCV. With the long-term use of HAART, HCV quasispecies have been increased in HIV / HCV co-infected patients.
